αMHC (α myosin heavy chain) directing expression to cardiomyocytes:

Using standard cloning techniques, the sensor and effector cassettes are placed under control of the minimal αMHC promoter, directing expression to cardiomyocytes.   The promoter is a 5.5kb fragment including the 3′ most exon of βMHC, the first three exons of αMHC (up to the start codon) as well as intervening sequences. HCN4 (hyperpolarization activated cyclic nucleotide-gated potassium channel) directing expression to the SA node and the AV node:

The sensor and effector cassettes are under control of the HCN4 promoter, directing expression to the SA node and the AV node.  Using homologous recombination in BAC RP23-281H22, the initiation codon of HCN4 was replaced with the expression cassette.  For construction details, click here.
